So I unpacked and assembled my own ISo of MGS4 and I cannot seem to locate it with the emulator.

I dumped the disc and made a digital file just like I have with other games from my PS 1 and 2, but when I go to my folder that should have my PS3 ISOs I find nothing and when I try to select that folder as my games folder for RPCS3 it gives me an "E boot failed *File path to my PS3 ISOs folder* " error.

Does this mean I assembled the ISO incorrectly? Or am I missing some step in putting the ISO into the emulator

Your ISO is probably still encrypted. You need to follow the instructions on the Quickstart Guide in order to dump your games properly.
